ABSTRACT

Abstract This case-control study evaluated the gene expression levels of interleukin (IL)-4, macrophage inflammatory protein type 1 alpha (MIP-1α), and metalloproteinase (MMP)-9, factors involved in the formation of giant cells in healthy peri-implant tissue and peri-implantitis. Thirty-five subjects (15 healthy and 20 with peri-implantitis), who met the inclusion and exclusion criteria, were included in this study. The peri-implant tissue biopsies were subjected to total RNA extraction, DNAse treatment, and cDNA synthesis. Subsequently, the reaction of real-time PCR was performed to evaluate the gene expression levels of IL-4, MIP-1α, and MMP-9 concerning the reference gene. IL-4 gene expression showed higher (18-fold) values in the Peri-Implantitis Group of Patients when compared with the Healthy (Control) Group (p<0.0001). Although MIP- 1α and MMP-9 gene expression levels were higher in diseased implants, they showed no significant differences (p=0.06 and p=0.2337), respectively. Within the limitations of this study, the results showed that in tissues affected by peri-implantitis, only levels of Il-4 were increased when compared with tissues in the control group.

ABSTRACT

Mycobacterium indicus pranii (MIP) earlier known as Mw is a soil-borne, non-pathogenic, saprophytic and rapidly growing strain of mycobacteria. MIP is approved as a vaccine/ immunomodulator for various indications including mycobacterium infections like leprosy in humans. Its administration has resulted in satisfactory clinical improvement, accelerated bacillary clearance, and increased immune responses to Mycobacterium leprae antigens, thereby shortening the full recovery time of the patients. It also shares its antigens with M.tuberculosis. In the last decade, RCTs have been done establishing immunotherapeutic properties of MIP in the treatment of leprosy, tuberculosis, warts and experimently in leishmaniasis. Through its immune inducing and cytotoxic property, it has also proved beneficial for human use especially in treating lung cancer. The beneficial role of it is also being explored in breast, cervical, oral, liver, and bladder cancers. Various studies on MIP have shown that it has immune-modulating properties in humans. The curiosity of the human mind has led to it being tried in Covid treatment trials. The results have shown that administering MIP has lowered inflammatory markers in Covid 19 patients, promising us for it to be a potential treatment option. More RCTs with a larger sample size should be done to establish this. Cytokine storm seen in bacterial sepsis is also decreased with MIP administration. Considering the encouraging results in hastening recovery in various diseases it appears that MIP is perhaps not being exploited to its fullest potential

ABSTRACT

Oncideres females girdle tree branches of the Fabaceae family, interrupting the sap flow and turning the wood conditions ideal for their larvae development. The bark of Stryphnodendron adstringens (Mart.) Coville, a species native to the Brazilian Cerrado, is widely used in the traditional medicine. The objectives were to report, for the first time, Oncideres saga (Dalman), using S. adstringens as a host and to describe the pattern of branch girdling and oviposition distribution by this insect on these branches. The diameter at the base and the length of the girdled branches were measured and the number of incisions made by the O. saga females to oviposit, per branch section (basal, median and apical), counted. The emerged specimens were counted and the diameter of the exit holes measured. The average diameter at the base of the girdled branches was 2.5 ± 0.16 cm and the length was 90.6 ± 4.6 cm. The average number of incisions per branch was 37.7 ± 2.7. Damage by O. saga can reduce the growth and cause losses on S. adstringens, a tree with great extractivism potential.

ABSTRACT

The objective of this work was to evaluate the topical, residual and agronomic effects of the insecticides Voliam Targo, Benevia, Lorsban 480 BR, Curbix 200 SC, Sperto, Verismo, Polytrin 400/40 CE, Curyom 550 EC, Polo 500 SC and Vertimec 18 EC in the control of the berry borer of coffee (H. hampei). The experimental design was completely randomized and four replicates were used in the trials. In the laboratory, a direct spray was sprayed on the insect (topical effect) and applied to the fruits of the coffee (residual contamination). In both experiments, each plot consisted of a Petri dish lined with filter paper, 10 fruits in the green stage and 10 adult females of the berry borer, originating from artificial breeding. Two field experiments were carried out at Campus Glória Experimental Farm. The first one was carried out with the objective of evaluating the residual effect of the insecticides on artificial infestation of the berry borer after applying the products to fruits at the beginning of maturation. The number of perforated fruits, dead females, number of eggs and larvae were evaluated in laboratory and field experiments. The second field experiment had as objective to evaluate the effectiveness in controlling the natural population of the berry borer. Two applications were performed with a 30-day interval, using a motorized turbocharger. The number of fruits bored in 50 fruits per plot and percentage of seeds bored in a sample of 250 seeds per replicate were evaluated. In the laboratory, all insecticides provided mortality higher than 80% in topical application and greater than 73% by residual contamination, except the insecticide Polo 500 SC, which provided a 55% mortality. In the field experiment with artificial infestation, all the insecticides differed from the control, maintaining residual control until 30 days after application and with mortality higher than 70%, reaching up to 100% of mortality. In the test with natural berry borer females infestation the insecticides differed from the control in all evaluated parameters, showed efficacy higher than 75% at 35 days after the second application. The insecticides Voliam Targo, Lorsban 480 BR, Curbix 200 SC, Sperto, Polytrin 400/40 CE and Verismo presented the best control results, being indicated for use in the management of the berry borer of coffee.

ABSTRACT

Chronic Obstructive Pulmonary Disease (COPD) is a common disease seen in Indian population. It is most commonly seen in the lower strata where beedi and cigarette smoking is most prevalent and starts at an early age. Smoke arising from cooking stoves during cooking is the cause of COPD in females. Though the number of males suffering from COPD is much larger than females, we observed a proportionate number of females in all age group suffering from this disease. It is the leading cause of mortality and should be studied in detail so as to avoid complications in such patients. We have studied the strength of respiratory muscles in such patients using newer techniques like Maximal Inspiratory Pressure (MIP) and 7 8 Maximal Expiratory Pressure (MEP). Terzano et al. (2008) and Khalil et al.(2014) have carried out similar studies in COPD patients. We have checked correlation between anthropometric measurements and strength of respiratory muscles, correlation between airway resistance and muscle strength and also compared the sensitivity and specificity of this technique using ROC (receiver operating Curve) as compared to the age old spirometry. These techniques may help in monitoring the status of COPD patients and assessing muscle weakness.

ABSTRACT

Objective: To investigate the effects of a recombinant endoglin-macrophage inflammatory protein 3α Fc-fusion protein (EM) vaccine on tumor angiogenesis and growth in mice with H22 hepatocellular carcinoma. Methods: An in vivo hepatoma mouse model was established. Seven days after subcutaneous inoculation of H22 tumor cells, mice were randomly divided into four groups: EM, endoglin Fc-fusion protein, macrophage inflammatory protein 3α Fc-fusion protein, and normal saline groups. Tumor volume and survival rate of mice were studied at 3-day intervals. Microvessel density of the tumors and tumor cell proliferation were detected by immunohistochemistry, and tumor cell apoptosis was detected by TdT-mediated biotinylated-dUTP nick-end label staining. The number of CD11c and CD86 positive dendritic cells were detected by flow cytometry. Results: Compared with the other groups, the tumor volume became smaller, and the survival time was longer in the EM-treated group. Besides, microvessel density and cell proliferation index were significantly lower, while the tumor cell apoptosis index was significantly higher in the EM-treated group. Besides the number of CD11c and CD86 positive dendritic cells in EM-treated mice was larger than that in other groups. Conclusions: EM Fc-fusion protein could effectively inhibit tumor growth through inhibiting endoglin-related tumor angiogenesis and cell proliferation, promoting tumor cell apoptosis, and could induce a certain degree of antitumor immune responses.

ABSTRACT

@#C-C motif Chemokine Ligand 3 Like 1 (CCL3L1) is characterized as a gene with copy number variable (CNV) and clustered at the segmental duplication on chromosome 17q12. CCL3L1 is responsible for the production of macrophage inflammatory protein (MIP) 1α that plays an important function in the immune system and host defense. Various copies range of CCL3L1 have been reported and associated with the diseases in different populations. Thus, this review aimed to summarise the distribution of CCL3L1 copy number from different populations according to the geographical region and highlighted the lacking of data from Malaysian population, which is one of the multi-ethnic countries due to the impacts of CCL3L1 copies on various diseases. Besides, we also outlined the methodologies available for the copy number typing. In overall, this review could provide significant information on the role of CCL3L1 copies in disease association and as well as providing evidence on the population diversity

ABSTRACT

Objective: To investigate the expression of IL-38 and MIP-2 in lung tissue of rats with pulmonary fibrosis induced by bleomycin, and to explore the significance of IL-38 and MIP-2 in pulmonary fibrosis in rats. Methods: 45 Wistar rats were randomly divided into saline control group ( group N), bleomycin group ( group B) and dexamethasone group ( group D) according to the random and control principle. On the 7 th, 14 th, and 28 th day, 5 rats were killed in each group. The pathological changes of lung tissue were observed by hematoxylin-eosin ( HE) staining in lung tissue. The expression of IL-38 and HYP in lung tissue of rats was measured by enzyme linked immunoassay ( ELISA) and the expression of MIP-2 in lung tissue of rats was measured by RT-PCR method. Results: (1) HE staining showed that the lung tissue from group B and group D developed from normal to inflammatory changes to pulmonary fibrosis. (2) The expression of IL-38 in group B and D decreased gradually, and the decrease was most obvious at 28 th day, which was lower than that in group N ( P<0. 05), and the expression of IL-38 in group B was lower than that in D group, and the difference was statistically significant ( P<0. 05). (3) The expression of MIP-2 and HYP increased gradually in group B and D, which were higher than those in group N, and the difference was statistically significant ( P<0. 05). The MIP-2 and HYP expressions in group B were higher than those of group D in the same period, and the difference was statistically significant ( P<0. 05). Conclusion: IL-38 and MIP-2 play an important role in the occurrence and development of bleomycin induced pulmonary fibrosis in rats. The application of dexamethasone can improve the degree of pulmonary fibrosis in rats. The effect may be related to the up-regulation of IL-38 and the downregulation of MIP-2.

ABSTRACT

The accessory pancreatic duct is the smaller and less constant pancreatic duct in comparison with the mainpancreatic duct. We investigated the patency of the accessory pancreatic duct and its role in pancreaticpathophysiology. The present study was carried out in the department of Anatomy and forensic medicine, ACSRGovt. medical college, Nellore, Andhra Pradesh, India and in the department of Anatomy, RIMS, Ongole, AndhraPradesh, India. With 96 human cadavers (64 males and 32 females) with 30 to 80 years age group have beenstudied after obtained of ethical committee permission. The dissection method was followed to obtain specimenof pancreas along with C-loop of duodenum, papillae were identified ad dye was injected into APD to see itspatency at MIP. 93.75% specimens present MIP. The prevalence of patency of APD at MIP in population understudy was 38.89%; this is more in males was 43.33%, when compared to the females was 30% but this differencewas not significant statistically. It observed that out of 35 patent APD cases, 33 cases had inter papillarydistance either 2cm or more than 2cm. I t indicates 94.29% of patent APD cases had inter-papillary distancee”2cm. So there is strong relationship between APD patency and inter papillary distance in population understudy. The means inter-papillary distance in patent APD cases was 2.29 ± 0.2cm and non-patent APD cases was1.85 ± 0.25 cm. This difference was statistically significant.

ABSTRACT

Objective Through the detection of MIP-1α and VEGF levels in serum and CSF of ALS patients, we evaluated the clinical value of MIP-1 and VEGF levels in ALS patients. Methods A total of 80 patients with ALS were collected from Kuangwu Hospital of Tongchuanin from Jan, 2012 to Jun, 2015, and 67 patients with non-inflammation neurological diseases were chosen as control group. We obtained CSF and serum samples, and the MIP-1α and VEGF levels were measured by ELISA method. Results The MIP-1α levels in serum and CSF of ALS patients were statistically and significantly higher than those in the control group (P<0.05) . The VEGF levels in serum of ALS patients were statistically and significantly higher than those in the control group (P<0.05).The VEGF levels in CSF of ALS patients were not higher than those in the control group, statistically insignificant (P>0.05).MIP-1 alpha and VEGF levels was positively correlated with ALS course. The MIP-1αand VEGF levels of the long duration group were greater than the short duration group (P<0.05) . Conclusion The rising of MIP-1α and VEGF may indicate an activation of compensatory responses in ALS which suggested that MIP-1 alpha and VEGF are involved in the pathophysiology of amyotrophic lateral sclerosis. We propose that MIP-1α and VEGF may be a useful biomarker witha prognostic and evaluating potential for ALS.

ABSTRACT

Objective: To understand the economic impact of over-the-counter medicines (OTC) medications on healthcare systems and the population in Europe and the Americas, with special focus in Brazil. Methods: Bibliographic review covering a 20-year timeframe. The articles were identified through the PubMed and Lilacs services, containing the terms "self-medication", "over-the-counter" and "savings", and their equivalents in Portuguese. Results: There is lack of publications focused on the economic impact that medicines can bring to the countries and the society. The most complete studies and statistics have been compiled by the three most important OTC manufacturers associations in the world and show an acceleration in switches and significant savings generated. In Brazil, a 2015 paper using conservative assumptions estimated a positive economic impact of OTC to the Brazilian Healthcare System (SUS) of R$ 364 million, about US$ 117 million. This is the only study that had the goal of an economic impact quantification conducted in the country. Conclusion: The new regulation RDC nº 98/2016 gives the possibility for the switch of many substances from prescription to OTC and it is imperative that robust studies are conducted to uncover all the impacts that can be expected, providing inputs for technical evaluations and even to attribute priority levels to them.

ABSTRACT

ABSTRACT: Pesticides are considered the first line of defense for the control of pests and diseases. At least in the short and medium term, the use of pesticides will remain an important strategy for pest management, allowing growers to produce crops of sufficient quality at low costs. A broad approach known as Integrated Pest Management (IPM) combines several different pest-control strategies, among which the combination of chemical and biological control stands out. It requires pesticides that achieve optimal control of target pests with minimal impact on the activity of biological control agents. Because of the dynamics of pest infestations, IPM routines are continuously adjusted by growers, requiring comprehensive information about pesticide effects on natural enemies. However, this information is not always available and often contradictory, which constrains the design of field recommendations. In this review, we focused on the importance of selective pesticides in IPM programs, and the effects of chemical pesticides on parasitoids, predators, and entomopathogenic fungi. We provided a detailed discussion of the challenges and constraints for research on pesticide effects on natural enemies, as well as for the resulting field recommendations.

ABSTRACT

Background: MIP is a cultivable, non-pathogenic organism, which shares several antigens with Mycobacterium tuberculosis and Mycobacterium leprae. It has several proposed clinical applications. However, its cytotoxic effect on pancreatic cancer has not been documented. Hence, the study was conducted to investigate MIP induced cytotoxicity on Mia-Pa-Ca2 cells. To determine the cytotoxic potential of heat killed Mycobacterium indicus pranii (MIP) on pancreatic cancer cells in vitro along with gemcitabine & 5-fluorouracil (5-FU). Mitogen-activated protein kinase (MAPK) level was also studied post MIP treatment. Methods: Cytotoxic effect of MIP, gemcitabine and 5-FU on Mia-Pa-Ca2 cells was determined. We have analyzed extent of apoptosis using flow cytometry and changes in p38 levels, c-Jun N-terminal kinases (JNK) and extracellular signalregulated kinase (ERK) using ELISA. Results: MIP not only exhibits cell cytotoxicity in dose dependent manner, but also enhances efficacy of gemcitabine and 5-FU when used in combination. Flow cytometry analyses reveals apoptosis of Mia-Pa-Ca2 cells post MIP treatment compared to untreated cells. MAPK pathway study using ELISA shows that p38 and JNK levels are suppressed while there is no change in ERK level. Conclusion: With these results we conclude that MIP is a cytotoxic agent. Cytotoxicity is exhibited by apoptosis. Combining MIP with gemcitabine and 5-FU shows synergistic effect (AU)

ABSTRACT

ABSTRACT: Genetic use restriction technologies (GURTs) were developed to preserve the intellectual property of genetically modified crops (GM) and ensure the return of investments made by industry to obtain technology delivered through seeds. The aims of this review are to discuss the GURTs and analyze their possible applications in integrated management of agricultural pests. There are two classes of GURTs: T-GURTs (trait-based GURTs), wherein the generated seed are viable, but the next generation does not express the trait of agronomic interest, and V-GURT (variety-based GURTs), in which plants produce non viable seeds. However, beyond the seed protection purpose, the GURTs could have also other application to solve agronomic problems. One of the most important is the use of GURTs as a tool to restrict gene flow of GM traits to relative weeds. In addition, it is proposed the use of this technology in integrated weed management by preventing the GMs seed germination, which produces volunteer plants that compete with the crop of interest. Also, these volunteer plants may serve as alternative hosts for insects and pathogens in between crop seasons. The GURTs could contribute to the control of undesirable agents in agricultural systems, reducing the use of pesticides and increasing crop yields.
